{"id":135,"date":"2023-10-26T11:31:08","date_gmt":"2023-10-26T15:31:08","guid":{"rendered":"https:\/\/projects.sjf.edu\/cangie\/?p=135"},"modified":"2023-10-26T11:31:08","modified_gmt":"2023-10-26T15:31:08","slug":"student-summer-research-charly-campanella","status":"publish","type":"post","link":"https:\/\/projects.sjf.edu\/cangie\/2023\/10\/26\/student-summer-research-charly-campanella\/","title":{"rendered":"Student Summer Research &#8211; Charly Campanella"},"content":{"rendered":"<p>Charly is a senior Biology student who is also pursuing minors in Chemistry and Psychology.\u00a0 This summer, she worked with Dr. Zachary Murphy through a sponsorship with the Summer Fellows Research Program on, &#8220;Investigating the Role of TET2 in Erythropoiesis with CRISPR\/Cas9,&#8221;<\/p>\n<p><span style=\"font-weight: 400\">Charly described her research: &#8220;<\/span><span style=\"font-weight: 400\">Erythropoiesis is a biological process that involves the production of erythrocytes, <\/span><span style=\"font-weight: 400\">commonly known as red blood cells. This process is crucial to human beings because <\/span><span style=\"font-weight: 400\">the molecules that interact with the red blood cells help to transport oxygen to all areas\u00a0<\/span><span style=\"font-weight: 400\">of the body. Red blood cells must take on a biconcave morphology so that the specific\u00a0<\/span><span style=\"font-weight: 400\">interactions between proteins and molecules can be made. This physical characteristic\u00a0<\/span><span style=\"font-weight: 400\">is created by the removal of the cell\u2019s nucleus and other cellular components during the\u00a0<\/span><span style=\"font-weight: 400\">developmental stage. If this developmental process is disrupted in any way, the cell will\u00a0<\/span><span style=\"font-weight: 400\">not be able to carry the normal amount of oxygen. These disruptions can lead to\u00a0<\/span><span style=\"font-weight: 400\">diseases such as anemia and myelodysplastic syndromes (MDS). One of the human\u00a0<\/span><span style=\"font-weight: 400\">genes, TET2, has been suggested to be related to MDS. It is thought that TET2 could\u00a0<\/span><span style=\"font-weight: 400\">play a role in the development of red blood cells, which could mean that a lack or\u00a0<\/span><span style=\"font-weight: 400\">mutation of the gene could lead to improper cells being produced. Our research has\u00a0<\/span><span style=\"font-weight: 400\">looked at this gene and what happens to cells when CRISPR\/Cas9 technology is\u00a0<\/span><span style=\"font-weight: 400\">introduced along with specific short guide RNA (sgRNA) sequences . The sgRNA\u00a0<\/span><span style=\"font-weight: 400\">sequences were designed to target exons 3, 7, and 11 of the gene. Our hypothesis is\u00a0<\/span><span style=\"font-weight: 400\">that if these three exons are targeted and mutated, then the cells will not take on the\u00a0<\/span><span style=\"font-weight: 400\">proper shape needed.<\/span><\/p>\n<p><span style=\"font-weight: 400\">&#8220;Alongside this idea, our research has also looked into finding inexpensive <\/span><span style=\"font-weight: 400\">alternatives to protocols and biochemical materials. Science can be quite costly, and\u00a0<\/span><span style=\"font-weight: 400\">this can create a financial barrier to students and schools that choose to work with\u00a0<\/span><span style=\"font-weight: 400\">erythroid research. We tested various protocols several times to determine whether or\u00a0<\/span><span style=\"font-weight: 400\">not these alternatives would be suitable to use for future experiments. This tied into the\u00a0<\/span><span style=\"font-weight: 400\">research above because this idea of finding alternatives came about while in the\u00a0<\/span><span style=\"font-weight: 400\">process of troubleshooting. Our goal is to create cost effective alternatives so that other\u00a0<\/span><span style=\"font-weight: 400\">institutions, like St. John Fisher, can conduct erythroid research.
